Dermatophytic onychomycosis (DO) is characterized as being a fungal infection of the nail. Infection can involve the nail unit (nail matrix, nail bed, or nail plate). Tavaborole, which was discovered and developed by Anacor Pharmaceuticals, is a topical antifungal from a novel class of antifungal agents known as benzoxaboroles that are being developed for the treatment of DO. Unlike the currently marketed therapies, Anacors tavaborole works by inhibiting a fungal enzyme, leucyl transfer RNA synthetase (LeuRS), which is required for fungal protein synthesis. The inhibition of protein synthesis terminates cell growth and promotes cell death, thereby eliminating the infection. Anacors proprietary boron-based product platform has also been used in developing a second product, AN-2718, which the company has stated will be used in the life-cycle management of its primary drug, tavaborole. Tavaborole has much higher bioavailability and efficacy than ciclopirox, the sole topical agent approved in the US market, because of increased nail penetration at almost 250-fold higher values.
