In this application note, different parameters were evaluated for optimizing oligonucleotide ion-exchange chromatography methods. For example, to help reduce carry-over, the study suggests increasing the initial concentration of salt, in this case NaCl. To help improve peak tailing, they investigated optimizing the mobile phase buffer as well as changing the counter ion. Overall, this work found that developing a separation with low carry-over and improved peak tailing of oligonucleotides can be achieved through the optimization of buffer concentration as well as choosing the correct counter ion when using YMC BioPro IEX QF anion exchange columns for the separation.
